Gene targeting strategy for B-hTLR2 mice. The exon 3 of mouse Tlr2 gene that encodes the extracellular domain was replaced by human TLR2 exon 1 in B-hTLR2 mice.
Strain specific TLR2 expression analysis in homozygous B-hTLR2 mice by flow cytometry. Peritoneal exudative macrophages (PEMs) were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hLTR2 mice (H/H). Mouse TLR2 was detectable in the wild-type C57BL/6 mice. Human TLR2 was exclusively detectable in the homozygous B-hTLR2 mice.
Strain specific analysis of TLR2 gene expression in WT and hTLR2 mice by RT-qPCR. Spleens were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hLTR2 mice (H/H). The mRNA expression of hTLR2 in B-hTLR2 mice (H/H) was less than those in the C57BL/6 mice (+/+), demonstrating that introduction of hTLR2 in place of its mouse counterpart changes the mRNA expression level of TLR2.
