Targeting strategy
Gene targeting strategy for B-hCXCL16 mice. The exons 2~5 of mouse Cxcl16 gene were replaced by human CXCL16 exons 2~5 in B-hCXCL16 mice.
mRNA expression analysis
Strain specific analysis of CXCL16 mRNA expression in wild-type C57BL/6 mice and B-hCXCL16 mice by RT-PCR. Lung RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-hCXCL16 mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human CXCL16 primers. Mouse Cxcl16 mRNA was detected in wild-type C57BL/6 mice. Human CXCL16 mRNA was detected in homozygous B-hCXCL16 mice but not in wild-type mice.
Protein expression analysis
Strain specific CXCL16 expression analysis in homozygous B-hCXCL16 mice by ELISA. Serum was collected from wild-type mice (+/+) and homozygous B-hCXCL16 mice (H/H), and analyzed by ELISA. Human CXCL16 was detectable in homozygous B-hCXCL16 mice.
