Gene targeting strategy for B-hCDCP1 mice.
The exons 2-7 of mouse Cdcp1 gene that encode the extracellular region were replaced by human CDCP1 exons 2-7 in B-hCDCP1 mice.
Strain specific analysis of CDCP1 gene expression in wild-type C57BL/6 mice and B-hCDCP1 mice by RT-PCR. Mouse Cdcp1 mRNA was detectable only in colon of wild-type C57BL/6 mice (+/+). Human CDCP1 mRNA was detectable only in homozygous B-hCDCP1 mice (H/H) but not in wild-type mice.
Strain specific CDCP1 expression analysis in homozygous B-hCDCP1 mice by western blot . Heart, liver and colon were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCDCP1 mice (H/H), and analyzed by western blot with species-specific anti-CDCP1 antibody. Human CDCP1 was detectable in colon of homozygous B-hCDCP1 but not in wild-type mice.
