Gene targeting strategy for B-hCD43 mice. The exon 3 of mouse Cd43 gene that encodes the extracellular domain was replaced by human CD43 exon 3 in B-hCD43 mice.
Strain specific analysis of CD43 gene expression in wild type (WT) and B-hCD43 mice by RT-PCR. Human CD43 mRNA was detectable only in splenocytes of the homozygous B-hCD43(H/H) mice but not in wild type (+/+) mice.
Strain specific CD43 expression analysis in WT and homozygous B-hCD43 mice by flow cytometry. Splenocytes were collected from wild type(+/+) and homozygous B-hCD43 (H/H) mice. Mouse CD43 was detectable only in the WT mice. Human CD43 was detectable only in the homozygous B-hCD43(H/H) mice.
