          • The NLRP3 gene encodes cryopyrin, a cytosolic protein featuring a nucleotide-binding domain and leucine-rich repeats (NLR), which is primarily expressed in white blood cells and chondrocytes. This protein regulates the assembly of the inflammasome and facilitates the cleavage of interleukin-1β (IL-1β), thereby playing a critical role in modulating immune responses to tissue injury, toxins, and infections
          • A LoxP-mouse Nlrp3 E4-10 minigene-STOP-LoxP chimeric was inserted between mouse Nlrp3 exon 3 and exon 4, with an alanine 350 to valine mutation in the subsequent mouse Nlrp3 exon 4. After mating with Cre mice, Cre-mediated recombination triggers a deletion event that excises the transcriptional termination signal cassette, thereby enabling expression of the point-mutated target gene.

          Targeting strategy

          Gene targeting strategy for B-Nlrp3*A350V flox mice. A LoxP-mouse Nlrp3 E4-10 minigene-STOP-LoxP chimeric was inserted between mouse Nlrp3 exon 3 and exon 4, with an alanine 350 to valine mutation in the subsequent mouse Nlrp3 exon 4. After mating with Cre mice, Cre-mediated recombination triggers a deletion event that excises the transcriptional termination signal cassette, thereby enabling expression of the point-mutated target gene.

          mRNA expression analysis

          Strain specific analysis of Nlrp3 mRNA expression in wild-type C57BL/6JNifdc mice and B-Nlrp3*A350V flox mice by RT-PCR. Spleen RNA was isolated from wild-type C57BL/6JNifdc mice (+/+) and B-Nlrp3*A350V flox mice (Mut/Mut). Mutated Nlrp3 mRNA was detectable in B-Nlrp3*A350V flox mice and in wild-type mice. The correct Nlrp3 sequence of the mice was confirmed by sanger sequencing and no leakage occurred without mating with Cre mice.
