Targeting strategy
Gene targeting strategy for B-hIGF2R mice. The extracellular, transmembrane and intracellular region of human IGF2R CDS were inserted into the mouse exon 3 in B-hIGF2R mice.
mRNA and protein expression analysis
- Strain specific analysis of IGF2R gene expression in wild-type mice and B-hIGF2R mice by RT-PCR. Mouse Igf2r mRNA was detectable in splenocytes of wild-type mice (+/+). Human IGF2R mRNA was detectable only in homozygous B-hIGF2R mice(H/H) but not in wild-type mice.
- Strain specific IGF2R expression analysis in homozygous B-hIGF2R mice by western blot. Heart tissue, kidney tissue and spleen tissue were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hIGF2R mice (H/H), and analyzed by western blot with anti-hIGF2R antibody. Human IGF2R was detectable in wild-type mice and homozygous B-hIGF2R mice, as the antibody is crossly reactive with IGF2R in human and mice.
