Gene targeting strategy for B-hCD52 mice.
The exons 1~2 of mouse Cd52 gene that encode the full-length protein were replaced by human CD52 exons 1~2 in B-hCD52 mice.
Strain specific analysis of CD52 gene expression in WT and B-hCD52 mice by RT-PCR. Mouse Cd52 mRNA was detectable in splenocytes of wild-type (+/+) mice. Human CD52 mRNA was detectable only in H/H, but not in +/+ mice.
Strain specific CD52 expression analysis in homozygous B-hCD52 mice by flow cytometry. Splenocytes were collected from WT and homozygous B-hCD52 (H/H) mice, and analyzed by flow cytometry with species-specific CD52 antibody. Mouse CD52 was detectable in WT mice. Human CD52 was exclusively detectable in homozygous B-hCD52 but not WT mice.
